It was understood the two men waited until customers were gone before allegedly kicking through the front door at about 7pm.
The 49-year-old staff member was closing the premises and quickly locked herself in the storeroom.
The men allegedly demanded money, however, the woman refused to leave the storeroom.
Police said the duo fled, taking the woman's keys and her Holden sedan from the club's car park.
"Although shaken, the woman was not injured during the incident and contacted police," a police spokesperson said.
"Officers from South Coast Police District attended a short time later and established two crime scenes, after the woman's Holden was found abandoned nearby on Acacia Place."
Club Dalmeny CEO Tony Casu was proud of the staff member's actions.
"She did an absolutely fantastic job, ticked all the boxes," he said.
"She was very shaken; we put her up in a motel for the night."
Mr Casu said the club was fully equipped with security cameras which will help police with their investigations.
